Facile Installation of Post-translational Modifications on the Tau Protein via Chemical Mutagenesis

چکیده

Post-translational modifications of proteins are ubiquitous in living organisms, as they enable an accurate control the interactions these macromolecules. For mechanistic studies, it would be highly advantageous to able produce vitro post-translationally modified with site-specificity. Here, we demonstrate one facile way achieve this goal through use post-translational chemical mutagenesis. We illustrate approach by performing site-specific phosphorylation and methylation tau, a protein that stabilizes microtubules whose aggregation is closely linked Alzheimer’s disease. then verify effects on ability tau microtubule polymerization, revealing particular unexpected role for at S199, which outside microtubule-binding region tau. These results show how mutagenesis present enables systematic analysis key involved pathogenesis

Post-translational modifications of protein biopharmaceuticals.

The majority of therapeutic proteins display one or more post-translational modifications (PTMs). These modifications normally influence the biochemical and therapeutic properties of such proteins. Choosing an expression system capable of generating an appropriate product PTM profile remains one of the most crucial decisions a drug developer must make. This review considers the PTMs most often ...

Lysine-Directed Post-translational Modifications of Tau Protein in Alzheimer's Disease and Related Tauopathies

Tau is a microtubule-associated protein responsible mainly for stabilizing the neuronal microtubule network in the brain. Under normal conditions, tau is highly soluble and adopts an "unfolded" conformation. However, it undergoes conformational changes resulting in a less soluble form with weakened microtubule stabilizing properties. Altered tau forms characteristic pathogenic inclusions in Alz...

Post-translational modifications of tubulin

Microtubules are the largest filamentous components of the eukaryotic cytoskeleton. In spite of their extraordinary level of structural conservation, microtubules fulfill a vast range of different functions in cells. How this functional diversity is achieved remains an open question; however, recent advances point towards post-translational modifications (PTMs) of tubulin as a potent mechanism ...
